Pathway-based polygene risk for severe depression implicates drug metabolism in CONVERGE

Background
The Psychiatric Genomics Consortium (PGC) has made major advances in the molecular etiology of MDD, confirming that MDD is highly polygenic. Pathway enrichment results from PGC meta-analyses can also be used to help inform molecular drug targets. Prior to any knowledge of molecular biomarkers for MDD, drugs targeting molecular pathways (MPs) proved successful in treating MDD. It is possible that examining polygenicity within specific MPs implicated in MDD can further refine molecular drug targets.
MethodsUsing a large case–control GWAS based on low-coverage whole genome sequencing (N = 10 640) in Han Chinese women, we derived polygenic risk scores (PRS) for MDD and for MDD specific to each of over 300 MPs previously shown to be relevant to psychiatric diagnoses. We then identified sets of PRSs, accounting for critical covariates, significantly predictive of case status.
ResultsOver and above global MDD polygenic risk, polygenic risk within the GO: 0017144 drug metabolism pathway significantly predicted recurrent depression after multiple testing correction. Secondary transcriptomic analysis suggests that among genes in this pathway, CYP2C19 (family of Cytochrome P450) and CBR1 (Carbonyl Reductase 1) might be most relevant to MDD. Within the cases, pathway-based risk was additionally associated with age at onset of MDD.
ConclusionsResults indicate that pathway-based risk might inform etiology of recurrent major depression. Future research should examine whether polygenicity of the drug metabolism gene pathway has any association with clinical presentation or treatment response. We discuss limitations to the generalizability of these preliminary findings, and urge replication in future research.
The onset of Arctic sea-ice snowmelt as detected with passive- and active-microwave remote sensing

Daily acquisitions from satellite microwave sensors can be used to observe the spatial and temporal characteristics of the Arctic sea-ice snowmelt onset because the initial presence of liquid water in a dry snowpack causes a dramatic change in the active-and passive-microwave response. A daily sequence of backscatter coefficient images from the NASA scatterometer (NSCAT) clearly shows the spatially continuous progression of decreasing backscatter associated with snowmelt onset across the Arctic Ocean during spring 1997. A time series of the active NSCAT backscatter and a scattering index from the passive Special Sensor Microwave/Imager (SSM/I) show similar trends during the time of the melt onset. An NSCATsnowmelt-onset detection algorithm is developed using the derivative of the backscatter with respect to time to select a melt-onset date for each pixel, generating a melt map for the Arctic sea ice. Comparison between this melt map and one previously generated from an SSM/I scattering index shows the NSCAT algorithm predicts the onset occurs 1−10 days earlier than the SSM/I-based algorithm for most portions of multi-year ice.

Effects of the imidazolinone herbicide AC243 997 [2-(5-isopropyl)-5-methyl-4-oxo-2-imidazolin-2-yl) nicotinic acid] on the growth of corn (Zea mays L. ‘Black Mexican Sweet’) cell suspension cultures were characterized. The herbicide caused half maximal growth inhibition at concentrations in the range of 10 nM to 30 nM. Significant reductions in protein synthesis were observed following treatment of cells with 10 μM herbicide. Analysis of free amino acid pool sizes indicated that levels of the biosyn-thetically related amino acids leucine and valine were substantially decreased in cells exposed to 10 μM herbicide. Supplementation of suspension culture growth media with leucine and valine plus isoleucine (1 mM each) reversed growth inhibitory effects of AC243 997 at levels ranging from 10 nM to 1 mM. The results suggest a specific interaction of this imidazolinone herbicide with leucine, valine, and isoleucine metabolism.

Objectives: This study examined whether children with distinct brain disorders show different profiles of strengths and weaknesses in executive functions, and differ from children without brain disorder. Methods: Participants were children with traumatic brain injury (N=82; 8–13 years of age), arterial ischemic stroke (N=36; 6–16 years of age), and brain tumor (N=74; 9–18 years of age), each with a corresponding matched comparison group consisting of children with orthopedic injury (N=61), asthma (N=15), and classmates without medical illness (N=68), respectively. Shifting, inhibition, and working memory were assessed, respectively, using three Test of Everyday Attention: Children’s Version (TEA-Ch) subtests: Creature Counting, Walk-Don’t-Walk, and Code Transmission. Comparison groups did not differ in TEA-Ch performance and were merged into a single control group. Profile analysis was used to examine group differences in TEA-Ch subtest scaled scores after controlling for maternal education and age. Results: As a whole, children with brain disorder performed more poorly than controls on measures of executive function. Relative to controls, the three brain injury groups showed significantly different profiles of executive functions. Importantly, post hoc tests revealed that performance on TEA-Ch subtests differed among the brain disorder groups. Conclusions: Results suggest that different childhood brain disorders result in distinct patterns of executive function deficits that differ from children without brain disorder. Implications for clinical practice and future research are discussed. (JINS, 2017, 23, 529–538)

Bacteriophytochromes (BphPs) are red-light photoreceptors found in photosynthetic and nonphotosynthetic bacteria that have been recently engineered as infrared fluorescent tissue markers. Light-induced, global structural changes are proposed to originate within their covalently bound biliverdin chromophore and propagate through the protein. Classical BphPs undergo reversible photoconversion between spectrally distinct light absorbing states, red (Pr) and far-red (Pfr), respectively. RpBph3 (P3), from Rhodopseudomonas palustris, photoconverts between a Pr and a unique near-red (Pnr) light-absorbing state. Due to size and photosensitivity of BphPs, structures of the intact proteins have not been resolved by nuclear magnetic resonance and/or X-ray crystallography. Therefore, structural details about the light and dark-adapted structures of the intact BphPs are not well understood at the molecular level. We have utilized fluid cell atomic force microscopy (AFM) to investigate the domain structure of intact P3 in its light-adapted state (Pnr). By varying the concentration of the protein, deposition time, and the ionic strength of the buffer, the aggregation of P3 on a mica surface can be controlled and single dimers may be observed in a biologically relevant media. Domain resolution has been achieved for several orientations of the dimer on the surface. The structural dimensions of the dimer have been compared to a modeled BphP in its intact form generated using PyMOL software. AFM experiments are currently underway to analyze the dark-adapted state (Pr) of P3 in order to observe the anticipated structural changes. Ultimately, the goal is to use AFM and other surface analytical methods such as scanning tunneling microscopy and electron microscopy to gain new insight into the unique photochemistry of P3.

Summary
Introduction
One of the hallmarks of the malignant phenotype is the ability of cells to grow in an autonomous manner with minimal cell death. This has been linked to diverse machinery of the proliferative and/or survival signaling pathways that become constitutively activated or deregulated in all human cancers. The proto-oncogenic driver mutations of Ras (H-, N-, K-ras, depending on tissue origin) or Raf gene are frequently tied to constitutive mitogen activated protein kinases (MAPK)/the extracellular signal regulated kinase (ERK1/2) signaling in a variety of tumors (Figure 9.1). Since the ERK signaling pathway is involved in both physiological and pathological cell proliferation, ERK1/2 inhibitors represent a desirable class of anti-neoplastic agents. Accumulating results have independently supported the promising anti-cancer effects of this class of inhibitor in numerous pre-clinical and clinical studies, i.e. CI-1040[1], selumetinib (AZD6244)[2–4], AS703026[5] and GSK1120212[6]. In fact, the MEK1/2 inhibitor CI-1040 (Pfizer) was the first signal transduction inhibitor tested in clinical trials in various advanced solid tumors[7]. Although no MEK1/2 inhibitors are approved for clinical use, kinase inhibitors that also inhibit Raf and VEGFR kinases (e.g. Sorafenib inhibiting multikinases)[8, 9] are successful anti-neoplastic agents against many types of cancer and FDA approved for the treatment of patients with unresectable hepatocellular carcinoma and advanced renal cell carcinoma [10, 11] (Figure 9.2). Such information support targeting this pathway, alone or in combination with other well-established growth and survival pathways, i.e. phosphoinositide 3-kinase (PI3K)/Akt/mTOR, STAT3, NFκB, heat shock proteins (Hsp90, Hsp70), histone deacetylases (HDAC), osteoblast (OB) as effective strategies across multiple cancers.

Myeloma is the second-most common hematologic cancer, accounting for approximately 1 percent of all cancers and 2 percent of all deaths from cancer. In recent years there have been major and significant advances in the diagnosis, imaging, molecular pathogenesis, and therapy of myeloma. This book describes these advances in a single, practical volume, placing them in the context of daily clinical practice. Different myeloma subtypes are discussed in individual chapters, integrating etiology, pathogenesis, molecular pathology, histology, and therapy in a single place for each condition. The book begins by discussing epidemiology, imaging techniques, pathogenesis, and other general considerations. INTRODUCTION
Despite recent advances in therapy, relapsed and refractory multiple myeloma (MM) remains a significant challenge and an area of unmet medical need. Median survival and responses to treatment are characteristically short. Relapsed and refractory MM is defined as patients who achieve at least a minor response (MR) or better followed by relapse and then progress on salvage therapy, or experience progression within 60 days of their last therapy. Successive treatment regimens typically result in progressively shorter response durations, which reflects emerging drug resistance. The observed decrease in response duration may also reflect changes in disease biology within each patient, with tumor cells expressing a more aggressive phenotype, higher proliferative thrust, and lower apoptotic rates.
Although several prognostic factors have been identified for newly diagnosed myeloma, factors that retain prognostic value in the context of relapsed/refractory disease remain to be comprehensively defined. Nonetheless, patients with poor risk include those with t(4;14) or t(14;16), deletion 17 or deletion 13, hypodiploidy, high- β2-microglobulin, light-chain disease, immunoglobulin A (IgA) isotype, and low serum albumin; clinical challenges in the relapsed/refractory population include renal failure, extramedullary disease, hyposecretory myeloma, and advanced bone disease.
The advent of novel therapies targeting disease biology and tumor microenvironment has significantly improved the prognosis for patients with relapsed and refractory disease. Bortezomib, a first-in-class proteasome inhibitor, and the immunomodulatory agents thalidomide (Thal) and lenalidomide now constitute “backbone” agents in this setting.
